How It Works

Experimental Design

Factor sensitivity is determined by testing serial dilutions of reference plasma (which contains approximately 100% factor activity) mixed with factor-deficient plasma (which contains 0% activity for the specific factor being studied). Each dilution represents a known proportion of reference plasma — for example, a 35% dilution contains 35% reference plasma and 65% factor-deficient plasma, resulting in approximately 35% factor activity.

Dilutions are expressed as percentages of reference plasma. The default study uses 8 dilution levels (25%, 30%, 35%, 40%, 45%, 50%, 60%, and 70%); the module supports custom dilution values and a configurable range of 3 to 15 dilutions. Each dilution is measured in up to 4 replicates, and the mean clotting time per dilution is used for regression analysis.

PT vs. aPTT pathway: The assay type (PT or aPTT) determines which coagulation pathway is being tested. PT factors include II, V, VII, and X. aPTT factors include VIII, IX, XI, and XII. The assay type is configured at experiment setup and affects the expected ULNR range.
